Somatic chromosomal changes of rat liver cells during in vitro cultivation.
نویسنده
چکیده
In order to examine how long diploid cells can be maintained in vitro cultivation without any chromosomal aberrations and to evaluate the mechanism of chromosomal heteroploid transformation, changes of chromosomes were studied in the course of serial in vitro transfers in four normal rat liver cell lines. As a result the diploid cells decreased in number gradually at early culture stage and disappeared completely in the periods between 350 and 500 days. The culture shifted to heteroploid as follow: diploidy—tpseudodiploidy—thypodiploidy—thypertriploidy or hypotetraploidy. This proces to heteroploid transformation is divided into five stages according to ploidy: 1and II-stages show diploidy, III-stage, pseudodiploidy, IVstage, hypodiploidy and V-stage, hypertriploidy or hypotetraploidy. Chromosomal heteroploid transformation and neoploastic conversion occurred in IV-stage of the cultures. A possible mechanism involved in the process of chromosomal transformation was discussed. ∗PMID: 4258082 [PubMed indexed for MEDLINE] Copyright c ©OKAYAMA UNIVERSITY MEDICAL SCHOOL Acta Med, Okayama 25, 87-100 (1971) SOMATIC CHROMOSOMAL CHANGES OF RAT LIVER CELLS DURING IN VITRO CULTIVATION

Background: Sperm chromatin insufficiencies leading to low sperm count and quality, infertility and transmission of chromosomal microdeletion and aneuploidies to next generations can be due to exposure to environmental pollutions, chemicals and natural or manmade ionizing radiation. In this project which has continued for more than 10 years and is unique in many technical aspects in Iran and in...
متن کاملEffects of 35 Hz, 2 mT Magnetic Field on Peripheral Blood Lymphocytes of Human In Vitro and Rat In Vivo
In this research we have studied the effects of extremely low frequency magnetic field on the peripheral blood lymphocytes chromosomes of human in vitro and rat in vivo. By this means, we used 60 blood samples of 10 men as well as 40 rats. Of 60 human blood samples ,40 samples (in two group of 20) were selected as test groups and placed into the 2 mT and 35 Hz magnetic field. The blood samples,...

Purpose: To evaluate the effects of lithium chloride on MSCs in vitro expansion rate. Materials and Methods: In this experimental study, bone marrow from 8 rats was plated at 5×105- cells/cm2 in the presence of 1,2,5,7 and 10 mM Lithium chloride and expanded through 3 passages. Twelve days after initiatial culture, the cells of different groups were stained with crystal violet in order to compa...

Background: Somatic cell nuclear transfer (SCNT) provides an appealing alternative for the conservation and improvement of genetic material of unique and endangered species. An important prerequisite for successful SCNT is the availability of good quality donor cells, as normal embryo development is dependent upon proper reprogramming of the donor genome so that embryonic genes can be appropria...
متن کاملBIO treatment enhances rat marrow-derived mesenchymal stem cell in vitro proliferation and viability
Introduction: Previous investigations have indicated that the presence of BIO (6-Bromoindirubin-3-Oxime) in medium of some cell culture enhances the cell proliferation and viability. The aim of the present study was to investigate the BIO effects on in vitro expansion of rat marrow-derived mesenchymal stem cells (MSCs) culture. Methods: In the present experimental study, bone marrow cells from ...
